摘要
微生物菌群的不同是影响人体舌苔表型的重要影响因素之一,实验利用PCR-DGGE法和测序法研究不同年龄段人体舌苔微生物菌群结构的差异。方法:分别刮取4个不同年龄段人群的舌苔,提取总DNA,PCR扩增后进行变性梯度凝胶电泳(DGGE)分析,分析不同年龄段人体舌苔微生物菌群结构的异同;并对差异条带进行测序分析,研究不同年龄段人群舌苔微生物群的差异。结果:韦永氏球菌属、普氏菌和链球菌等是各年龄段均有的舌苔菌群,产气荚膜梭菌多为儿童舌苔上携带,Uncultured prevotellaceae是老年舌苔特有菌群,普雷沃氏菌属是青年中年舌苔特有菌群。结论:不同年龄段人体舌苔上的微生物菌群有一定的差异,本实验结果可以为临床上舌苔微生物的理论研究和诊断提供参考。
Different microbe groups are important factors affect human tongue coating, here, PCR- DGGE method were used to detect human tongue coating structure of microbial flora. Methods : tongue coating of four age groups were sampled respectively, and the DNA of microbes in coating on the tongue were extracted and purified ;16S rDNA were amplified by polymerase chain reaction (PCR) ; and finally the PCR product were analysed by the denaturing gradient gel electrophoresis (DCCE) ; the specific bind were sequenced and anlysed by BLAST. Resuh:Wepericorum,Pricococcus and Strep- tococcus are the tongue coating flora of all ages. Clostridium perfringens are carried on the tongue of Most of the of children, Unccultured prevotellaceae bacterium B00064B52 were found only on the old group,and the genus Poulet was discoveried in young and middle-aged group. Conclusion: there are certain differences in microbial flora on the tongue fur of different age groups. The results of the ex- periment can provide reference for the theoretical study and diagnosis of the tongue coating microorganism.
