新生大鼠缺氧缺血再灌注损伤后ROCK-2,α-SMA与ROCK信号通路的关系 被引量:2

Relationship between ROCK-2, α-SMA and ROCK Signaling in neonatal rats after hypoxic-ischemic reperfusion injury

摘要 目的检测α-SMA(α-Smooth muscle actin)、ROCK-2(Rhoass ociated coiled-coil forming protein kinase-2)在缺氧缺血新生大鼠脑组织的表达,探讨Rho(Rho guanosinetriphosphatases)/ROCK信号通路的激活在新生大鼠缺氧缺血再灌注脑损伤中的意义。方法将SD大鼠的7 d龄新生鼠(n=48)随机分为两组:真手术组包括3个不同时段的缺氧缺血性脑病后2 h、6 h、24 h组和相对时段的假手术对照组,采用多克隆抗体免疫组织化学SP法观察α-SMA、ROCK-2的表达。结果假手术组在各时段α-SMA、ROCK-2表达无明显变化;HIBD组中α-SMA,ROCK-2在不同时间点呈上升趋势;ROCK-2与α-SMA的表达呈正相关。结论 ROCK-2与α-SMA在新生大鼠缺氧缺血性脑损伤后随着时间表达呈上升趋势,证明ROCK-2与α-SMA参与HIBD的病理生理过程,从而说明RHO/ROCK信号通路的激活参与了新生鼠缺氧缺血性脑病的发生发展。 Objective To detect the expression of α-SMA(α-Smooth muscle actin) and ROCK-2(Rhoass ociated coiled-coil forming protein kinase-2) in the brain tissue of neonatal rats with hypoxia-ischemia, and to explore the significance of activation of the Rho(Rho guanosinetriphosphatases)/ROCK signaling pathway in hypoxic-ischemic reperfusion injury in neonatal rats. Methods 7-day-old neonatal rats(n=48) from SD rats were randomLy divided into two groups: the real surgery group consisted of 2, 6, and 24 hours after hypoxic ischemic encephalopathy and the sham-operated control group. The expression of α-SMA and ROCK-2 was observed by polyclonal antibody immunohistochemical SP method. Results There was no significant change in the expression of α-SMA and ROCK-2 in the sham operation group. The α-SMA and ROCK-2 in the HIBD group showed an increasing trend at different time points. ROCK-2 was positively correlated with the expression of α-SMA. Conclusion ROCK-2 and α-SMA increase with time after hypoxic-ischemic brain damage in neonatal rats, which proves that ROCK-2 and α-SMA participate in the pathophysiological process of HIBD, thus indicating the activation of RHO/ROCK signal pathway is involved in the development of hypoxic ischemic encephalopathy in neonatal rats.
