饮食干预对碘乙酸所致大鼠骨性关节炎的辅助改善作用研究 被引量:1

摘要 目的:观察饮食干预对碘乙酸诱导的大鼠骨性关节炎模型的辅助改善作用。方法:32只雄性SD大鼠按体重随机分为4组。除正常对照组(NG)外,其余各组给予高脂饲料,碘乙酸注射法建立骨性关节炎模型。连续饲养4周后,将饮食干预组(DIG)大鼠给予普通饲料,模型对照组(MG)与药物治疗组(DTG)仍给予高脂饲料; DTG与DIG膝关节腔内注射透明质酸钠,连续4周。每周称量大鼠体重;检测各组大鼠空腹血脂水平、膝关节影像学与病理学变化、血清MMP-13、Aggrecan、Col II的含量。结果:相比于NG,MG、DTG与DIG的体重增长较快;造模4周后,血清TC均有差异(P <0. 05),DTG与DIG血清TG均有差异(P <0. 05);饮食干预4周后,相比于DTG,DIG大鼠体重、血清MMP-13含量显著下降(P <0. 05),Col-II、Aggrean含量显著升高(P <0. 05),膝关节影像学上损伤与光镜下病理改变均得到有效改善。结论:饮食干预对碘乙酸诱导的大鼠骨性关节炎有较好的辅助治疗作用。 [Objective] To observe the Auxiliary effect of Dietary intervention on Rat Osteoarthritis induced by iodoacetic Acid. [Method] According to body weight, 32 male SD rats were randomly divided into 4 groups with 8 rats in each group. In addition to the normal group (NG), the rest groups were fed with high-fat diet, and the model was established by injecting iodacetic acid into knee joint cavity bilaterally. After 4 weeks of continuous feeding, the rats in the dietaryintervention group (DIG) were fed with common diet, while the model group (MG) and the drug treatment group (DTG) were still fed with high-fat diet. The DTG and the DIG were injected sodium yaluronate into the knee joint cavity for 4 weeks. The weight of the rats was measured weekly. Blood lipids levels, the imaging changes and the pathological changes of knee joint the content of MMP-13、Aggrecan and Col II in serum were measured in rats of each group. [Result] Compared tothe NG, the DTG and the DIG gained weight faster, there were significant differences in serum TC levels between the MG, the DTG and the DIG( P〈0.05); and therewere significant differences in serum TG levels between the DTG and the DIG after 4 weeks modeling ( P 〈0.05). Compared to the DTG, the body weight and the content of serum MMP-13 of the rats in the DIG was significantly decreased (P 〈0.05), and the contents of Col-II and Aggrean increased significantly ( P〈0.05). The imaging injury of knee joint and the pathological changes under light microscope were all improved effectively. [Conclusion] Dietary interventionhas a good auxiliary effect on rat osteoarthritis induced by iodoacetic acid.
