电针远端取穴对神经根型颈椎病模型大鼠镇痛相关因子影响 被引量:2

Effects of Electroacupunctural Distal Acupoints on Analgesic Factors of Cervical Spondylosis Radiculopathy Model’s Rats

摘要 目的:观察电针远端取穴对神经根型颈椎病(CSR)模型大鼠IL-1β、IL-6的影响,探讨电针远端取穴对CSR模型大鼠的镇痛机制。方法:SPF级Wistar大鼠随机分为3组:空白组、模型组、电针组,每组各20只。除空白组除外,余两组采用手术方法将尼龙渔线放至大鼠C6~7、T1神经根腋下造成CSR模型。电针组大鼠取双侧"太冲""合谷"电针,每天1次,每次20 min。干预7 d后检测各组大鼠机械痛阈。采用ELISA法、Real-Time PCR法分别检测压线处脊髓和神经根组织IL-1β、IL-6含量及IL-1βmRNA、IL-6 mRNA表达。结果:模型组痛阈较空白组显著降低(P〈0.05);干预后电针组痛阈较模型组显著升高(P〈0.05)。模型组IL-1β、IL-6含量及其mRNA含量较空白组显著升高(P〈0.05);电针组较模型组IL-1β、IL-6含量及其mRNA含量显著降低(P〈0.05)。结论:电针对CSR模型大鼠具有镇痛作用,通过降低脊髓及神经根组织IL-1β、IL-6含量和mRNA表达可能是其镇痛机制之一。 Objective:To observe the effects of electroacupunctural distal acupoints on analgesic factors of cervical spondylosis radiculopathy model's rats,and to discuss its analgesic mechanism. Methods:Randomly divided SPF Wistar rats into 3 groups,blank group,model group,and electroacupuncture group(n=20). Excepted the blank group,put nylon fishing line into rats' C6-7,T1 nerve roots with operational method to simulate CSR model. Treated electroacupuncture group's rats with electroacupuncture at bilateral LR3 and LI4,20 minutes each time,once a day. Measured mechanical pain threshold of 3 groups of rats after 7-day-treatment. Detected contents of IL-1β,IL-6 in spinal cord and nerve tissue by ELISA,and determined espression of IL-1βmRNA,IL-6 mRNA by Real-Time PCR. Results:Mechanical pain threshold significantly decreased more in the model group than in the blank group(P〈0.05). After treatment,electroacupunture group got a more significantly increase in mechanical pain threshold than model group(P〈0.05). Compared with the blank group,contents of IL-1β and IL-6 as well as espressions of IL-1βmRNA and IL-6 mRNA in spinal cord and nerve tissue increased significantly in the model group(P〈0.05). Compared with the model group,contents of IL-1β and IL-6 as well as espressions of IL-1βmRNA and IL-6 mRNA decreased significantly in the electroacupuncture group(P〈0.05). Conclusion:Electroacupunctural distal acupoints impacts an analgesic effect on CSR-model's rats. Electroacupuncture treatment can reduce contents of IL-1β and IL-6 as well as espressions of IL-1βmRNA and IL-6 mRNA in rats' spinal cord and nerve tissue,it may be one of its analgesic mechanism.
