五味降压散对自发性高血压大鼠降压、利尿作用实验探究 被引量:2

摘要 目的:实验探究五味降压散对自发性高血压大鼠(Spontaneously hypertensive rats,SHR)是否有利尿作用。方法:14周龄SHR大鼠随机分为模型组、氢氯噻嗪组(噻嗪组)和五味降压散组(五味组),每组10只。10只WKY大鼠作为正常对照组。五味降压散以3.2 g·kg^-1的浓度灌胃给药,连续给药12周;给药期间动态观察4组大鼠的收缩压(SBP)、24 h尿量、24 h尿蛋白含量(TP)变化。每两周固定时间测定大鼠血压1次以观测药物降压效果;每两周做1次代谢笼收集各组大鼠24 h尿液,观察尿量并用双缩脲法做24 h尿蛋白定量(TP)测定。结果:五味组给药0-11周SHR尿量有增加趋势,给药12周时,尿量较模型组明显增加,差异有统计学意义(P〈0.05)。五味组在给药4周、6周时TP值明显高于正常组,差异有统计学意义(P〈0.05);12周时TP值明显低于模型组,差异有统计学差异(P〈0.05)。结论:五味降压散对自发性高血压大鼠具有一定的利尿作用。 Objective:To verify the diuretic effect of Wuwei Jiangya San(WWJYS)on spontaneous hypertensive rats(SHR). Methods:The 14-week-old spontaneous hypertensive rats were randomly divided into model group,hydrochlorothiazide group(thiazine group)and WWJYS group(Wuwei group),10 in each group,and 10 WKY as a control group. WWJYS was administered by intragastric administration at a concentration of 3.2 g·kg^-1. The systolic blood pressure(SBP),24 h urine volume and 24 h urine protein content(TP)of the four groups were observed dynamically during continuous administration. Rat blood pressure was measured once every two weeks to observe the antihypertensive effect of the formula. The metabolic cage was used every two weeks to collect the urine of each group for 24 hours. The urine volume was observed and the 24-hour urine protein quantitation(TP)was determined by the biuret method. Results:The urine volume was significantly increased in the Wuwei group at 0-11 weeks compared with the model group and the difference was statistically significant(P〈0.05)at 12 weeks. The TP value was significantly higher than the control group at 4 weeks and 6 weeks(P〈0.05),and significantly lower than the model group at 12 weeks(P〈0.05). Conclusion:WWJYS has a certain diuretic effect on spontaneous hypertensive rats.
