树突状细胞亚型变化在儿童咳嗽变应性哮喘中的临床意义 被引量:1

摘要 目的:观察咳嗽变异性哮喘(coughvariantasthma,CVA)儿童外周血树突状细胞m DCs和pDCs两种亚型的变化,并探讨其临床意义。方法:收集海口市第三人民医院2017年3月至2018年4月收治的C VA儿童6 2例。另取同期健康体检的儿童4 0例作为对照。用流式细胞仪检测外周血m DCs和p DCs的比例,ELISA法检测外周血血浆中IFN-γ和IL-4表达。结果:对照组和CVA组儿童m DCs间差异无统计学意义(P>0.05),CVA组儿童p DCs为(0.92±0.24)%,显著高于对照组的(0.31±0.08)%(P<0.01),m DCs/p DCs比值在CVA组儿童为0.62±0.15,显著低于对照组的1.68±0.34(P<0.01)。CVA组儿童IFN-γ和IFN-γ/IL-4比值分别为(40.47±8.45)ng/mL和0.92±0.14,显著低于对照组的(53.36±10.42) ng/mL和2.51±0.46(P<0.05和P<0.01)。CVA组儿童IL-4为(43.81±7.16)ng/mL,显著高于对照组的(21.27±4.15)ng/mL(P<0.01)。结论:CVA儿童外周血pDCs显著升高,导致了患儿体内的Thl/Th2失衡,pDCs有可能成为CVA诊断和治疗的新靶点。 Objective: To detect the changes of mDCs and pDCs subtypes in peripheral blood of children with cough variant asthma(CVA). Methods: Sixty-two CVA and 40 healthy children were enrolled in this study. The mDCs and pDCs was detected by flow cytometry. IFN-γ and IL-4 was detected by ELISA analysis. Results: There was no significant difference of mDCs between CVA and healthy children(P〈0.05). The pDCs was(0.92±0.24)% in CVA children, which was higher than that of(0.31±0.08)% in healthy children(P〈0.01). The mDCs/pDCs was 0.62±0.15 in CVA children, which was lower than that of 1.68±0.34 in healthy children(P〈0.01). The IFN-γ and IFN-γ/IL-4 was(40.47±8.45) ng/mL and 0.92±0.14 in CVA children, which was lower than that of(53.36±10.42) ng/mL and 2.51±0.46 in healthy chi ldren(P〈0.05 and P〈0.01). The IL-4 was(43.81±7.16) ng/mL in CVA children, which was higher than that of(21.27±4.15) ng/mL in healthy children(P〈0.01). Conclusion: The pDCs in peripheral blood is increased significantly, which causes Thl/Th2 imbalance in CVA children.
