摘要
目的:观察和解肝脾腹部推拿法对非酒精性脂肪肝(NAFLD)大鼠肠黏膜组织MLCK信号通路的调控作用。方法:采用高脂饮食喂养10周建立NAFLD大鼠模型,再以和解肝脾腹部推拿法治疗4周后对比手法干预组和模型对照组体质量、肝湿重变化,用Western blot方法,检测手法干预前后,NAFLD大鼠模型肠黏膜组织MLCK及其介导的PMLC蛋白表达的变化,利用异硫氰酸荧光素-葡聚糖(FITC-Dextran)示踪法检测干预前后大鼠肠黏膜通透性变化。结果:与模型对照组相比较,手法干预组大鼠体质量及肝湿重明显降低(P 〈0. 05);手法干预组肠黏膜组织MLCK蛋白表达及其介导的P-MLC蛋白表达较对照组明显降低(P 〈0. 05);手法干预组大鼠肠袋外漏出FITC-Dextran含量与模型对照组相比明显降低(P 〈0. 05)。结论:和解肝脾腹部推拿法可降低NAFLD大鼠体质量与肝湿重,其通过调控肠组织MLCK信号通路,改善肠上皮屏障功能,降低肠道黏膜的通透性,有利于阻断LPS等毒素对肝脏的侵袭,此可能为和解肝脾腹部推拿法治疗NAFLD的作用机制。
Objective: To observe the effect of"regulating the function of liver and spleen in abdominal massage"on NAFLD regulation of MLCK signal pathway in rats intestinal mucosa. Methods: NAFLD rat model was established by feeding high fat diet for 10 weeks,and abdomen kneading method for 4 weeks. Then Intervention group and model control group's body weight and liver wet weight were compared. Then Western blot method was used to detect changes of expression of MLCK and its mediated P-MLC protein in NAFLD rats before and after intervention and FITC-Dextran changes of intestinal permeability of rats were detected before and after intervention. Results: Compared with the model control group,body weight and liver wet weight of the intervention group decreased significantly(P〈0.05). The expression of MLCK in the intervention group was significantly lower than that in the control group(P〈0.05),and the expression of P-MLC protein in the intervention group was significantly lower than that in the control group(P〈0.05). Manipulation intervention group's content of FITC-Dextran was significantly lower than that of the control group(P〈0.05). Conclusion: Abdomen kneading method can reduce body weight andrat liver wet weight of NAFLD rats,through the MLCK signaling pathway in intestinal tissue,improve the intestinal epithelial barrier function,reduce intestinal mucosal permeability,and it is beneficial to block the invasion of LPS and other toxins to the liver which may be the mechanism of reconciliation and abdominal massage in the treatment of NAFLD.
