氧化苦参碱缓释片抗血栓作用的研究 被引量:3

The study on the antithrombotic effects of oxymatrine sustained-release tablets

摘要 目的探讨氧化苦参碱(oxymatrine,OMT)缓释片对急性血瘀模型大鼠血液流变学及血栓形成的影响。方法将120只Wister大鼠,随机分为6组,即正常对照组、模型组、OMT200mg·kg^(-1)组及OMT缓释片50、100、200mg·kg^(-1)三个剂量组,每组20只。采用皮下注射肾上腺素加冰水冷浴法制备大鼠急性血瘀模型,灌胃给药,观察OMT缓释片对急性血瘀模型大鼠全血及血浆粘度、体内血栓形成时间、体外血栓长度及干、湿质量、血小板聚集率、凝血时间(PT、APTT)指标的影响。结果 OMT组及OMT缓释片100、200mg·kg^(-1)组均能明显降低血瘀大鼠血液粘度、延长体内血栓形成时间,缩短体外血栓长度,降低血栓的干、湿质量,降低血小板聚集率、延长凝血时间。结论OMT缓释片能明显改善急性血瘀模型大鼠血液流变性指标,抑制血栓生成,且作用结果优于OMT组。 Objective To investigate the effect of oxymatrine(OMT)sustained release tablets on hemorheology and thrombosis in rats with acute blood stasis.Methods A total of 120 Wistar rats were randomly divided into 6 groups,namely normal control group,model group,OMT 200 mg·kg^-1 group,and OMT sustained-release tablets groups(50,100,200 mg·kg^-1).The acute blood stasis model was established by subcutaneous injection of adrenaline and ice-cold bath.and intragastric administration.Rats were orally administered with saline or OMT respectively.Several parameters were detected to investigate the treatment effects of OMT sustained-release tablets on acute blood stasis model rats,including blood viscosity,in vivo thrombosis time,in vitro thrombus length and mass,platelet aggregation rate and coagulation time.Results OMT and OMT sustained release tablets(100 and 200 mg·kg^-1)significantly reduced the blood viscosity,length of extracorporeal thrombus,dry and wet mass of thrombus,and platelet aggregation rate;in the meanwhile,prolonged thrombosis time,and clotting time.Conclusion OMT sustained-release tablets could attenuate the alteration of hemorheology and formation of thrombus in rats with acute blood stasis.Moreover,OMT sustained-release tablets showed better therapeutic effects than OMT.
