摘要
目的:探讨针刺对骨骼肌拉伤后纤维化进程的抑制效应及可能机制。方法:224只雄性SD大鼠随机分为对照组、拉伤组、针刺组和拉伤后针刺组。每组又分为即刻、2、4、7、14、21和28 d组,每组8只。对照组无干预;拉伤组用大鼠腓肠肌拉断仪建模;拉伤后针刺组在拉伤后每隔天针刺1次;针刺组与拉伤后针刺组同步。干预末次取腓肠肌备用。采用Masson染色法观察肌纤维损伤及炎症、Western blot检测TGF-β1、CTGF、MMP-1、TIMP-1蛋白的表达。结果:与对照组相比,拉伤组骨骼肌TGF-β1,CTGF,MMP-1,TIMP-1不同时相起蛋白表达均出现显著增加(P <0. 05,P <0. 01);与拉伤组相比,拉伤后针刺组骨骼肌TGF-β1,CTGF,TIMP-1不同时相起蛋白表达均出现显著减少(P <0. 05),MMP-1无显著变化(P> 0. 05)。拉伤组MMP-1/TIMP-1蛋白比值仅在2 d、4 d明显高于对照组(P <0. 05),之后迅速恢复(P> 0. 05);拉伤后针刺组MMP-1/TIMP-1蛋白比值7 d起持续明显高于拉伤组(P <0. 01)。结论:针刺可能通过下调TGF-β1/CTGF通路、上调MMP-1/TIMP-1蛋白比值抑制骨骼肌损伤修复中纤维化的发生发展。
Objective: To discuss the inhibitory effect and mechanism of acupuncture on fibrosis in skeletal muscle recovery process. Methods: Totally 224 male Sprague Dawley rats were randomly divided into control group,muscle strain group,acupuncture group and acupuncture after muscle strain group. And then each group was divided into immediately,2 days,4 days,7 days,14 days,21 days and 28 days subgroups depending on the sampling time points. There were 8 rats in each subgroup. The control group had no intervention; the rats of muscle strain group were established the injury model by gastrocnemius rupture instrument; rats of acupuncture after muscle strain group were needled once every other day after the strain,the rats of acupuncture group were needled at the same time. The gastrocnemius was collected after experiment. The muscle fiber injury and inflammation were observed by Masson staining; TGF-β1,CTGF,MMP-1 and TIMP-1 expressions were tested by Western blotting.Results: Compared with control group,the expressions of TGF-β1,CTGF,MMP-1 and TIMP-1 in muscle strain group increased at all times( Ps 〈0. 05); compared with muscle strain group,the expressions of TGF-β1,CTGF and TIMP-1 in acupuncture after muscle strain group decreased at all times( Ps 〈0. 05) while MMP-1 had no change( P〉 0. 05). The ratio of MMP-1/TIMP-1 of muscle strain group was higher than that of control group at2 d and 4 d groups( P〈 0. 05),and then it was recovered quickly( P〉 0. 05). The ratio of MMP-1/TIMP-1 of acupuncture after muscle strain group was higher than that of muscle strain group from the 7 th day( P 〈0. 05).Conclusions: Acupuncture may inhibit the occurrence and development of fibrosis in skeletal muscle repair by down-regulating TGF-β1/CTGF pathway and up-regulating the ratio of MMP-1/TIMP-1 protein.
