从江香猪和大白猪不同组织/器官中GHR、JAK2、STAT3基因的表达分析 被引量:4

摘要 为了分析从江香猪和大白猪不同组织中GHR、JAK2、STAT3基因的表达差异,试验取6月龄、体况相近且饲养环境相同的贵州从江香猪和大白猪各4头,分别提取心脏、肝脏、肺脏、肾脏、脂肪、大肠、小肠和背最长肌组织的总RNA,以猪GAPDH基因为内参,设计GHR、JAK2、STAT3基因的荧光定量引物,以荧光定量PCR法检测GHR、JAK2、STAT3基因在两个品种不同组织中的表达量。结果表明:GHR、JAK2、STAT3基因在两个品种不同组织/器官中均有表达,其中GHR基因在肝脏中相对表达量最高,且与其他组织/器官相比差异极显著(P〈0. 01),在脂肪中相对表达量最低; JAK2基因在肝脏中相对表达量最高,且与其他组织/器官相比差异极显著(P〈0. 01),在背最长肌中相对表达量最低; STAT3基因在肾脏中相对表达量最高,且与其他组织/器官相比差异极显著(P〈0. 01),在背最长肌中相对表达量最低。说明GHR、JAK2、STAT3基因在动物生长发育过程中发挥重要作用,在不同组织器官中的表达存在差异。 The aim of the present study was to analyze the expression difference of GHR, JAK2 and STAT3 gene in different tissues of Congjiang xiang pig and large white pig. Four 6-month-old Guizhou Congjiang xiang pig and four large white pigs with similar body condition and same feeding environment were selected. Total RNAs of heart, liver, lung, kidney, fat, large intestine, small intestine and longissimus dorsi muscle tissue were extracted from the pigs. The fluorescent quantitative primers were designed for GHR, JAK2 and STAT3 genes, and the porcine GAPDH gene was used as internal reference. The relative expression levels of GHR, JAK2 and STAT3 genes in different tissues or organs of the two varieties were detected by fluorescence quantitative analysis. The results showed that GHR, JAK2 and STAT3 genes were expressed in dif- ferent tissues of two varieties. The relative expression level of GHR gene in liver was the highest, the difference was significant compared with other tissues or organs ( P〈0. O1 ), and the relative expression level was the lowest in fat. The relative expression level of JAK2 gene was the highest in liver and the difference was significant compared with other tissues or organs( P〈0.01 ), and the relative expression level was the low- est in longissinms dorsi muscle. The relative expression level of STAT3 gene was the highest in kidney, the difference was significant compared with other tissues or organs (P〈0. 05), and the relative expression level was the lowest in longissimus dorsi muscle. The results indicate that GHR, JAK2 and STAT3 genes play an important role in the growth and development of animals and their expressions are different in different tissues or organs.
