猴头菌浸膏多糖及蛋白质含量测定方法的比较 被引量:8

摘要 为了更好地控制猴头菌浸膏的质量,试验分别采用3,5-二硝基水杨酸法、蒽酮-硫酸法和苯酚-硫酸法测定猴头菌浸膏中多糖的含量,分别采用福林酚法和考马斯亮蓝法测定猴头菌浸膏中蛋白质的含量,从中筛选出适合猴头菌浸膏的多糖及蛋白质含量的测定方法。结果表明:用蒽酮-硫酸法测定多糖含量时,结果高于其他两种方法,且稳定性、精密度、加标回收率、重复性的相对标准偏差(RSD)均小于5. 00%;用福林酚法测定蛋白质含量时,结果高于考马斯亮蓝法,且稳定性、精密度、重复性及加标回收率的RSD均小于5. 00%。说明蒽酮-硫酸法操作简便、稳定可行,是最适合测定猴头菌浸膏中多糖含量的方法;福林酚法操作简便、稳定可行,是最适合测定猴头菌浸膏中蛋白质含量的方法。 The aim of the present study was to better control the quality of Hericium erinaceus extractum. The contents of polysaccharides in Hericium erinaceus extractum were determined by 3,5-dinitrosalicylic acid method,anthrone-sulfuric acid method and phenol-sulfuric acid method,respectively. And the content of protein in Hericium erinaceus extractum was determined by Folin phenol method and Coomassie brilliant blue method. Then the suitable method for the determination of polysaccharides and protein in Hericium erinaceus extractum was selected. The results showed that the content of polysaccharides determined by anthrone-sulfuric acid method was higher than that by the other two methods,and the relative standard deviation(RSD) of stability,precision,repeatability and recovery rate of the method were all less than 5. 00%. The content of protein determined by Folin phenol method was higher than that by Coomassie brilliant blue method. And the RSD of stability,precision,repeatability and recovery of standard addition were all less than 5. 00%.The results indicated that the method of anthrone-sulfuric acid was simple,stable and feasible,and it was the most suitable method for the determination of polysaccharides in Hericium erinaceus extractum. The method of forint phenol was simple,stable and feasible,and it is the most suitable method for the determination of protein in Hericium erinaceus extractum.
