正交设计法优化壮药蛇尾草中毛蕊花糖苷提取工艺及其含量动态积累分析

Optimization of the extraction technology of verbascoside from Pogonostemon auricularius(Linn.) Hassk.by orthogonal test and research on its dynamic content accumulation

摘要 目的:优选蛇尾草中活性成分毛蕊花糖苷最佳提取工艺,并测定其含量动态积累过程,考察蛇尾草药材最佳采收时间。方法:采用高效液相色谱法,根据正交设计表考察甲醇浓度、溶剂用量、提取时间和提取次数对毛蕊花糖苷含量测定结果的影响;对不同采收时间的蛇尾草药材进行含量测定,对其含量变化进行分析。结果:最终确定优化提取方案为70%甲醇,溶剂容量为25mL,超声提取30min,提取1次;蛇尾草中毛蕊花糖苷的含量6月时到达峰值,随后逐渐降低,¨月采收药材含量最低。结论:所得优化工艺操作简单合理,结果准确。蛇尾草中毛蕊花糖苷含量6月份最高,但其最佳采收期的确定需进一步结合药效开展研究。 Objective: To optimally select the best extracting technology of verbaseoside in ogonosternon auricularius (Linn.) by orthogonal test, and to study dynamic change of content of verbaseoside in Pogonostemon auricularius (Linn.) Hassk. between different growth periods and to search the best harvest time. Methods : The orthogonal experiment method was used for studying the effect of methanol concentration, solvent dosage, times and duration extraction were evaluated. Then the content of verbaseoside in Pogonostemon auricularius (Linn.) Hassk. harvest in different growth periods was determined and analyzed by HPLC on the basis of the best extracting technology. Results: The best extraction process was refluxing for 45 minutes with 25 mL 70% methanol for I time. The content of verbaseoside in Pogonostemon auricularius (Linn.) Hassk. tend to peak in June and then reduce to the lowest in November. Conclusion: The established extraction was stable and reliable. And the max content of verbaseoside in Pogonostemon auricularius (Linn.) Hassk. is in June, but the determination of the optimum harvest time should be further combined with pharmaeodynamie studies.
