摘要
目的 研究 P5 3、P16和 c- erb B- 2蛋白表达与原发性肝细胞癌生物学行为的关系。方法 应用免疫组化链霉素抗生素蛋白——过氧化物酶连结法 (L SAB)检测 6 0例原发性肝细胞癌和癌旁肝组织 P5 3、P16和 c- erb B- 2蛋白表达。结果 P5 3、P16和 c- erb B- 2在原发性肝细胞癌中的阳性率分别为 4 1.6 %、35 %和 5 8.33% ;癌旁肝组织的阳性率分别为 8.33%、88.33%和 2 1.6 7% ,差异有显著性 (P<0 .0 1或 P<0 .0 5 )。 P5 3和 c- erb B- 2与肝细胞癌的病理组织学分级具有一定程度的相关性 ,P16蛋白与病理组织学分级有显著差异 (P<0 .0 5 )。P16、c- erb B- 2和 P5 3均与转移有关。P5 3蛋白表达与 c- erb B- 2蛋白表达具有显著的协同性 (P<0 .0 5 ) ,与 P16蛋白表达呈负相关性 (P<0 .0 5 )。结论 肿瘤的多因素分析比单因素分析更有价值 ,癌基因 c- erb B- 2和抑癌基因 P5 3、P16蛋白的表达异常及协调作用在原发性肝癌的发生、发展中起重要作用。
Objective To study the relationship between the expression of P53?P16 and c erbB 2 proteins and biologic behavior of hepatocellular carcinoma(HCC).Methods The expressions of P53?P16 and c erbB 2 proteins in tumor cells and peritumor tissues in 60 cases of HCC were detected by using immunohistochemistry labelled straptavidin biotin(LSAB)method.Results Positive rates of P53?P16 and c erbB 2 proteins were 41.67%?35% and 58.33% in HCC group and 8.33%?88.33% and 21.67% in pericancerous group respectively.They showed a significant difference( P < 0.01 or P <0.05).The level of the expression of both P53 and c erbB 2 oncoprotein in HCC showed some relationship to the pathological grades.The positive expression of P16 was related to the pathological grades( P <0.05).P16?c erbB 2 and P53 gene expressions were related to the metastasis.The expression of P53 protein had significant synergy with the expression of c erbB 2 protein( P <0.05),but was negatively correlated with the expression of P16 protein( P <0.05).Conclusion The analysis of multiple oncogene and/or antioncogene is more valuable than that of signle oncogene and/or antioncogene.The altered expression of oncogene c erbB 2 and antioncogenes P53 and P16 might play a promotive role in the tumorigenesis and development of hepatocellular carcinoma.
