中药对兔晶状体上皮细胞凋亡的cAMP的抑制作用 被引量:6

摘要 目的 :观察中药复明眼液对凋亡的兔晶状体上皮细胞 (lensepithelialcells,LEC)的环磷酸腺苷 (cAMP)的影响及其作用机制。方法 :采用终浓度 2 5 0 μmol/LH2 O2 诱导体外培养的兔LEC凋亡。用放射免疫分析法检测不同浓度的复明眼液对凋亡的兔LEC的cAMP含量的影响。结果 :各实验组兔LEC的cAMP含量 ,模型组为 (89.2± 10 .2 )pmol/g ,空白组为 (6 0 .6± 10 .7)pmol/g ,两者比较差异有非常显著性 (P <0 .0 1) ,说明兔LEC凋亡时 ,cAMP浓度明显增高。中药治疗C组 [(81.3± 11.2 )pmol/g]与模型组比较差异没有显著性 (P>0 .0 5 ) ,说明终浓度为 10 0 μg/ml的复明眼液对兔LEC的cAMP含量没有明显作用。中药治疗D组 [(6 9.5± 11.8)pmol/g]与模型组比较差异有显著性 (P <0 .0 5 ) ,说明终浓度为 2 0 0 μg/ml的复明眼液可以降低兔LEC的cAMP含量。中药治疗E组 [(5 2 .2± 7.6 )pmol/g]和F组 [(4 1.5± 8.1)pmol/g]分别与模型组比较 ,差异均有非常显著性 ,显示终浓度为40 0 μg/ml和 80 0 μg/ml的复明眼液 ,有显著减少兔LEC中cAMP含量的作用。上述各中药治疗组 ,随着复明眼液浓度的提高 ,对兔LEC的cAMP作用愈强 ,即呈一定的量效关系。结论 :终浓度 2 5 0 μmol/L的H2 O2 Objective:To investigate the effect of traditional Chinese medicine FMYY on cAMP in rabbit LEC apoptosis and explore the possible mechanisms involved.Methods:Apoptosis of rabbit LEC cultured in vitro was induced with H 2O 2 with a final concentration of 250 μmol/L. Radioimmunoassay(RIA) was used to examine the effects that different amounts of FFMY had on cAMP concentration in rabbit LEC apoptosis.Results:There was a significant difference in cAMP concentrations between all experimental groups[(89.2±10.2)pmol/g] and the control group[(60.6±10.7)pmol/g](P<0.01),indicating that cAMP concentration greatly increases in LEC apoptosis. There was no significant difference between traditional Chinese medicine treatment in group C[(81.3±11.2)pmol/g] and the control group(P>0.05),indicating that the final dose of 100 μg/ml FMYY had no obvious effects on cAMP concentrations in LEC apoptosis. There was a significant difference between traditional Chinese medicine treatment in group D[(69.5±11.8)pmol/g] and the control group(P<0.05),indicating the final dose of 200 μg/ml FMYY can reduce LEC cAMP concentrations. Compared with the experimental group,traditional Chinese medicine treatments in group E[(52.2±7.6)pmol/g] and group F[(41.5±8.1)pmol/g] showed significant differences,indicating the final concentrations of 400 μg /ml and and 800 μg/ml FMYY can significantly reduce LEC cAMP concentrations. The traditional Chinese medicine concentrations mentioned above have obvious effects on cAMP concentrations in rabbit LEC apoptosis and there was a dose-dependent effect when elevated concentrations were administered.Conclusion:H 2O 2 in a final concentration of 250 μmol/l can induce rabbit LEC apoptosis,accompanied by an increase in cAMP in rabbit LEC apoptosis. Traditional Chinese medicine FMYY can decrease cAMP concentrations in apoptotic rabbit LEC,so it appears to protect against the apoptosis of rabbit LEC cultured in vitro when induced by H 2O 2.
