实验性矽肺大鼠免疫细胞功能的研究 被引量:1

Study on Functions of Immune Cells in Rats with Expenmental Silicosis

摘要 我们用二氧化硅悬液给大鼠气管内注射,制备了大鼠实验性矽肺模型。在注射后6月,其肺部出现肉眼可见的明显病变,肺泡巨噬细胞数量比对照大鼠之值明显增加。几项免疫细胞功能的检测结果表明,与正常大鼠比较,矽肺大鼠肺泡巨噬细胞对SiO_2的刺激产生白细胞介素-1(IL-1)增加,脾细胞及其分泌免疫球蛋白的细胞(IgSC)的数量增加,然而脾细胞对ConA和LPS的增殖反应无明显差别。根据这些结果可以认为,在气管内注入了SiO_2的大鼠,肺泡巨噬细胞数量及其产生IL-1的增加属于局部的原发免疫学变化;而脾细胞总数及其中IgSC数量的增加继发性变化,此过程可以用来作为对一些矽肺病人出现自身抗体原因的解释。 Six weeks after intratracheal injection,of silica suspension to rats,grosspathological lesions werc seen and the number of alveolar macrophages(AM) increased in lungs of silica-treated rats as compared with that ofthe untreated.The results of immune cell assays showed that both theIL-I production by AM and the number of immunoglobulin secreting cells (IgSC) of spleens were higher in silica-treated rats than in controls.Based on these results,the increase in the number of and IL-I produc-tion by AM may be interpreted as primary alteration in silica-inhaled animals,while the elevated numbers of total spleen cells and of IgSC in spleen can be regarded as secondary changes.Because increase in polyclonal IgSC may give rise to auto-antibodies in vivo,it is not hard to un-derstand why anti-nuclear antibody,rheumatoid factor or other auto-antibodies appear frequently in some silicotic patients.
