NGF和MK801对Aβ_(25-35)诱导海马细胞损伤的干预作用

摘要 目的 探讨神经生长因子(NGF)和MK801对Aβ_(25-35)的细胞毒性的干预作用。方法 将培养7天的3天内新生大鼠海马细胞分别加入NGF(10ng/ml)、MK801(10μg/ml)、等体积培养液前后,加入b-淀粉样蛋白_(25-35)(Aβ_(25-35),10μg/ml),采用β-tublin、MAP2免疫组化鉴定是否为结构完整的海马神经元,MTT检测细胞活力,以及ChAT免疫组化检测免疫阳性细胞数目。结果β-tublin、MAP2免疫组化证实所培养的为特异性结构完整的海马神经元。MTT及免疫组化检测显示Aβ_(25-35)导致细胞活力显著降低及ChAT免疫阳性细胞明显减少,NGF、MK801可有效预防Aβ_(25-35)诱导的神经毒作用(P<0.01),以及可轻度改善Aβ_(25-35)诱导的神经毒作用,但无统计学意义(P>O.05)。结论 NGF和MK801能有效地阻止和轻度地修复Ab_(25-35)诱致的海马细胞损伤,对阿尔茨海默病的防治提供了一定的实验依据。
