生精细胞生长因子纯化及其活性检测

SGF PURIFICATION AND ACTIVITY DETECTION

目的:研究生精细胞生长因子(SGF)提取纯化方法。方法:取兔睾丸研碎、匀浆,通过盐析、透析和SephadexG-100层析,制成SGF粗品,再过DEAE-纤维素阴离子交换柱纯化,获得SGF纯品。结果:SGF粗品分离获得α、β、γ3种蛋白质,其中γ型蛋白质是SGF纯品,对生精细胞生长有明显促进作用,有特异性,能促进细胞的DNA合成。结论:SGF纯品是一种有活性的特异性蛋白,其分子量为10KD。

Objective: Methodological Study on Purification and abstract of Sperm-growth factors (SGF). Method:Rabit testicles was levigated into serous fluid, crude SGF was made through salting out,dialysis and chromatogra phy for the fluid. Purified SGF was made by Purification of the crude Product through DEAE -fibrinous ariion exchange Column. Results: Crude SGF was seperated into three proteins α, β and γ, which Protein u is a purified SGF and can facilitate growth of spermatogenie cells through promotion of DNA synthesis. Conclusion: Purified SGF is a active and specific protein with molecular weight 10 KD.
