摘要
HC108细胞系为贴壁生长具有上皮细胞形态特征的分化程度较高的人肝癌细胞。AFP分泌阴性,细胞平均倍增时间为32.21小时,细胞分裂指数为27.75‰,软琼脂克隆形成率为5.0%,能形成裸鼠移植瘤,支原体为阴性。染色体和流式细胞术分析为超二倍体核型,17号染色体改变最为明显。
A new human hepatocellular carcinoma cell line from surgical hepatocellular carcinoma tissue of a patient has been established. The morphological changes of the cell was examined under light and electron microscopy. The growth curve and cloning efficiency of the cultured HC-108 Cells were investigated. The chromosome and cell cycle were analyzed by G-banding and flow cytometry. The tumorigenesis on nude mice and mycoplasma were studied. The epithelial cell line has not secreted alpha - fetoprotein. A doubling time of 32. 21h in medium RPMI-1640 supplemented with 10% CS and 5.0% clonal efficiency in soft agar were observed. When they were transplanted subcutaneously into the back of athymic nude mice(balb/c,nu/nu) .tumors were formed at the sites of inoculation,which were shown to be hepatocellular carcinoma,similar in morphology to the original tumor from which they were derived. HC- 108 cells had a chromosome mode of 80 - 100 with one identifiable marker.
