血塞通对局灶性脑缺血细胞凋亡及Bax,Bcl-2表达的影响 被引量:3

摘要 目的 :探讨血塞通对局灶性脑缺血大脑皮质神经细胞凋亡及Bax ,Bcl- 2基因表达的影响 .方法 :采用DNA原位末端标记TUNEL技术及ABC免疫组织化学方法 ,检测了局灶性脑缺血后 6h大鼠大脑皮质的凋亡神经元数及Bax ,Bcl- 2的平均灰度 .结果 :血塞通处理组神经细胞凋亡数 2 2 5± 6 3较缺血组 41 5±5 5明显减少 (P <0 0 5 ) .而Bax ,Bcl- 2的平均灰度 95 4± 4 9,77 2± 2 1亦较缺血组 82 5± 6 6 ,97 3± 4 4增加和减少 (P <0 0 5 ) .结果提示 :血塞通处理有效减少皮质凋亡神经细胞数 ,而此变化可能与凋亡基因Bax ,Bcl- In order to explore the change of number on neuronal apoptosis and expression of Bax, Bc1-2 in cerebral cortex following ischemia, the rats were performed local ischemia operation derived from middle cerebral artery block. Rats both Xuesetong treatment group and ischemia group were sacrificed and the cerebral cortex was taken. The tissues were sectioned into 20μm frozen sections. The sections were stained by DNA in situ terminal labeling technique (TUNEL) and ABC immunohistochemiscal method by antibody of Bax and Bc1-2. The number of apoptosis neurons and average gray degree for Bax, Bc1-2 was counted in each group. Results: The number of apoptosis neurons in Xuesetong group decreased greatly than that of ischemia group,, while the gray degree for Bax and Bc1-2 in Xuesetong group increased and decreased than that of ischemia group respectively. It showed that Bax expression was downregulated, but expression of Bc1-2 was upregulated following Xuesetong administration. Conclusion: Treatment of Xuesetong exerts protective effects on cerebral cortex neuron following ischemia.
