局灶性脑缺血后不同时相大鼠大脑皮层神经细胞凋亡的变化

Change of Number on Neuronal Apoptosis in Cerebral Cortex in Different Time Following Local Ischemi

目的 :探讨局灶性脑缺血不同时相大鼠大脑皮质神经细胞凋亡的变化 .方法 :采用DNA原位末端标记TUNEL技术 ,检测了局灶性脑缺血后 1,3,6 ,12h大鼠大脑皮质的凋亡神经元数 .结果 :正常组及缺血后 1,3,6 ,12h神经元凋亡数分别为 7± 2 ,37± 7,49± 5 ,6 3± 7,89± 7,表明缺血随时间延长 ,凋亡神经元数进行性增多 (P <0 0 5 ) .结论 :缺血可致大脑皮质神经元凋亡 。

In order to explore the change of number on neuronal apoptosis in cerebral cortex following ischemia, the rats were performed local ischemia operation derive from middle cerebral artery block. Rats were sacrificed at 1?h, 3?h, 6?h, 12?h following ischemia. The cerebral cortex was taken and sectioned into 20?μm frozen section. The sections were stained in situ terminal labeling technique (TUNEL). The number of apoptosis neurons was counted in each group. Results: The number of apoptosis neurons in control and 1?h, 3?h, 6?h, 12?h following ischemia is respectively 7±2, 37±7,49±5,63±7,89±7. It showed that apoptosis was trigged by ischemia in cerebral cortex. The number of apoptosis gradually increases related to the ischemia time.
