切断部分背根神经对脊神经节和脊髓II板层一氧化氮合酶表达的影响

摘要 目的 :观察部分背根神经切断对猫脊神经节 (SG)和脊髓II板层一氧化氮合酶 (NOS)表达的影响 .方法 :采用尼克酰胺嘌呤二核苷酸脱氢酶 (NADPH -d)法显示单侧部分背根切断 (切断一侧L1~L5,L7~S2 背根节 ,保留L6为备用根 )后 10d的猫L6SG和L6脊髓II板层NOS的分布及变化 .结果 :NOS的阳性物主要分布于SG胞体偏小的神经元和脊髓II板层的神经膨体内 .部分背根切断后 10d ,手术侧SG内胞体偏小的NOS阳性神经元数量及II板层NOS阳性神经膨体密度均较非手术侧者增多 (P <0 0 5 ) .结论 :部分背根切断不仅致备用SG胞体偏小的神经元NOS表达增加 ,且II板层NOS的阳性膨体数量亦有增多 .提示由NOS产生的一氧化氮 (NO) Objective: To explore the effect of partial dorsal root nerve rhizotomy on the expression of nitric oxide synthase (NOS) in spinal lamina II and spinal ganglion (L 6). Method: The change of NOS in spinal lamina II and SG (L 6) on 10 days following unilateral partial dorsal root rhizotomy (L 1~L 5, L 7~S 2 dorsal roots were sectioned, but L 6 dorsal root was kept) was studied with a denosime dinucleotide phosphate diaphorase (NADPH-d). Results: the positive reaction of NOS was mainly distributed in small and medium sized neurons of SG and varicosities of lamina II. However, following operation, the number of positive neurons in SG and the density of varicosities apparently increased than that of un-operated side (P<0.05). Conclusion: Partial dorsal root nerve rhizotomy leads to the upregulation of expression of NOS in SG and spinal lamina II. It indicated that nitric oxide(NO) derived from NOS play role in the plasticity of the spinal cord.
