摘要
目的 :为比较不同组织切片方法对免疫组织化学结果的影响 .方法 :用成年SD大鼠L5脊髓分别制成冰冻 (2 0 μm)和石蜡 (5 μm)切片 ,用兔抗BDNF多克隆抗体作ABC免疫组化染色 .结果 :冰冻切片组背角Ⅱ板层除有大量BDNF阳性神经膨体外 ,尚可见一些阳性神经元 .石蜡切片组则仅有大量阳性神经膨体 ,而几乎未见阳性神经元 .结论 :不同制片法对免疫组织化学结果有影响 ,即冰冻切片可能较石蜡切片具有更高的免疫反应敏感性 .本文中石蜡切片较低的BDNF免疫活性可能与其制作过程中有机溶剂 (酒精、二甲苯 )的使用和
To assess the different influences of frozen and paraffined sections on the results of immunohistochemistry, the L 5 segments of spinal cords of adult Sprague-Dawley rats were taken and made into frozen (20?μm in thickness) and paraffined (5?μm ) sections respectively. The two sorts of sections were all stained using immunohistochemistry ABC method. Results: in the group of frozen sections, the lamina II of spinal dorsal horn abounded in BDNF positive varicosities. Additionally, some positive neurons of BDNF could also be observed. In contrast, positive neurons could hardly be seen in the group of paraffined sections. Our results indicate that frozen sections may have stronger immunoreactivity than paraffined ones. The weak immunoreactivity of paraffined sections possibly attributes to the loss of BDNF caused by the use of organic solutions and /or by the heating when sections were embedded with paraffin in the course of making paraffined sections.
