摘要
目的:测定小鼠血浆及各组织中羟基喜树碱HCPT浓度。方法:先用氢氧化钡-硫酸锌沉淀样品中蛋白,再用荧光分光光度法测定其含量。结果:HCPT在激发光波长为365nm、发射光波长为430nm、狭缝为10nm时荧光强度最大;HCPT浓度在0.01~0.5μg/mL范围内与荧光强度有良好的线性关系;平均回收率为血浆78.0%,肝80.1%,肺89.6%,胃72.6%,肠72.8%,CV均低于10%。结论:方法简便、经济、灵敏、准确,适用于血浆及组织中HCPT浓度的测定。
Objective:Assay the concentration of hydroxycamptothecine(HCPT)in plasma and tissues of mouse.Methods :Proteins in the samples were precipitated by barium hydroxide -zinc sulpate first.Then assay its content by fluo-rospectrophotometry.Results:When excitation wavelength is365nm,emission wavelength is430nm,narrow is10nm,HCPT reaches its maximum fluorescent intensity.Within the range of 0.01~0.5μg /mL,the concentration of HCPT has good linearity with fluorescent intensity.Average recover rate:plasma 78.0%,live80.1%,lung89.6%,stomach72.6%,intestines72.8%,all CV are lower than10%.Conclusion:The method is simple,economical and accurate,suitable for the assay of concentration of HCPT in plasma and tissues.
