摘要
用5%Tween-20处理油松针叶类囊体,将基粒类囊体的基粒片层和间质类囊体间质片层的侧端部位(margins部位)膜(curvedmembrane)“切”下来,把散开的基粒片层和margins部位光合膜中镶嵌的PSⅠ大分子颗粒离心分离,并分别进行SDS电泳,分离获得镶嵌在油松光合膜不同区域中的PSⅠ大分子两条电泳带。对两条电泳带中PSⅠ大分子分别进行光吸收特征的物理测定,确定出两条PSⅠ电泳带的光吸收特征曲线基本一致〔1~3〕。同时,将SDS电泳分离得到的PSⅠ两条带中PSⅠ大分子分别进行各自结合脂类的TLC分析测定。得出油松针叶类囊体的光合膜中,随着PSⅠ大分子复合颗粒在类囊体光合膜中镶嵌区域的不同,结合在PSⅠ大分子上的脂类数量和种类有所不同。
After the thylakoid be treated with 5% Tween 20 we have gotten the appressed granal membranes and the non appressed ganal granal margins membranes of the thlakoid of Chinese Pine ( Pinus tabulae formis) leaf′s chloroplast by centrifugalizing. Then PS Ⅰ complexes located in the appressed granal membranes and non appressed granal margins be separated two protein complexes by SDS PAGE. In our experimental result there is different numbers and kinds of the lipids of bounded on two protein complexes of PS Ⅰ located in the appressed granal membranes and non appressed granal margins.
