摘要
目的:研究D-半乳糖(D-Galactose,D-gal)对大鼠海马细胞衰老相关信号的影响。方法:取6月龄雄性SD大鼠30只,平均分成两组,每组15只,D-gal组利用进行D-gal连续腹腔注射(120mg/kg.day),对照组注射同体积的PBS共10周。利用Western blot和免疫组织化学观察SirT1和FoxO3a的表达变化。结果:Western blot和免疫组化结果显示,SirT1在D-gal组表达明显降低(分别为p=0.02,p=0.018);FoxO3a的免疫组化检测发现,D-gal组的海马锥体细胞FoxO3a在胞核的分布明显高于对照组(p=0.002);而相对胞浆的分布却显著降低(p=0.005)。结论:短期给予D-gal能够导致大鼠海马衰老信号的改变。
Objective: To study the senescence-related cell signaling alteration of D-galactose induced ageing rat's hippocampus.Method: 6-month of age SD male rats were applied for experiment.Rats of D-gal group(n=15) were given D-gal(120mg/kg.d) every day by intraperitoneal injection for 10 weeks and rats in control group(n=15) were given 0.1M PBS for 10 weeks.Western blot and immunohistochemistry were applied to detect SirT1,FoxO3a.Results: SirT1 attenuated showed by western blot and immunohistochemistry in D-gal group.(p=0.02,p=0.018 respectively).In pyramidal cells of D-gal induced hippocampus,nuclear localization of FoxO3a was higher than that of control(p=0.002);However,cytoplasm translocation decreased in D-gal group than that of control hippocampus(p=0.005).Conclusion: Senescence cell signaling changes induced by short-term D-gal injection of rats.
