山羊不同成熟阶段卵母细胞玻璃化冷冻敏感性的探讨

摘要 试验比较了山羊GV期和MII期卵母细胞对玻璃化冷冻损伤敏感性的差异。卵巢取自昆明当地屠宰场,将采集的GV期卵母细胞分成两组。第Ⅰ组直接用于毒性实验和玻璃化冷冻保存;第Ⅱ组经体外成熟至MII期后用于毒性实验和玻璃化冷冻。冷冻方法采用Cryoloop法。结果表明:毒性实验组和冷冻保存组卵母细胞的形态正常率和发育率均显著低于新鲜对照组(P<0.05)。毒性实验组GV期卵母细胞的形态正常率显著低于MII期(P<0.05),但是二者卵裂率之间差异不显著(P>0.05)。解冻后GV期卵母细胞的形态正常率显著低于MII期卵母细胞((60.37±2.91)%vs(82.91±3.01)%,P<0.05),而且解冻后GV期卵母细胞的卵裂率也显著低于MII期卵母细胞((42.81±2.94)%vs(57.91±1.06)%,P<0.05)。但是解冻后GV期和MII期卵母细胞的囊胚发育率之间差异不显著((8.51±1.46)%vs(12.41±3.74)%,P>0.05)。进一步表明,GV期未成熟卵母细胞对玻璃化冷冻的敏感性要明显高于MII期成熟卵母细胞。
