摘要
通过大鼠脑落体致伤模型,放免法检测伤后5、15、60及180分钟鼠脑皮质和血浆血栓素 B_2(TXB_2)与6-酮-前列腺素 F_(12)(6 Keto-PGF_(1α)含量,同时行病理学研究,探讨 TXA_2和 PGI_2与继发性脑损害的关系。结果:伤后3小时内,脑损伤皮质和血浆中 TXB_2及6-Keto-PGF_(1α)含量逐渐增高,TXB_2/6-Keto-PGF_(1α)比值亦逐步递增;病理学检查证实,随访后时间延长,脑损害程度愈加重。提示 TXA_2与 PGI_2代谢失衡是引起脑损伤早期继发性脑损害加重的因素之一,而测定脑组织和血浆中 TXB_2与6-Keto-PGF_(1α)含量可反映继发性脑损害的程度。
Correlation of secondary brain injury withTXA_2 and PGI_2 levels following head injurywere studied in 94 male Wistar rats.TXB_2 and6-Keto-PGF_1 in the quick frozen damagedcortical tissue and plasma were determined byRIA.The spreading of Evans blue in corticaltissue,light and electron microscopic studywere also undertaken.The results showed thatthe concentration of TXB_2 and 6-Keto-PGF_1 wasprogressively increased after cerebralcontusion both in the damaged tissue and inplasma,and the ratio between TXB_2 and 6-Keto-PGF_1 increased during the early stage afterinjury.Meanwhile,the tissue damage was moreaggrevated.It is concluded the content ofTXB_2 and PGI_2 is increased gradually followingcerebral injury and the imbalance between TXAand PGI_2 may be one of the important factorsin the development of secondary brain injury.
