摘要 目的:建立固相萃取-分光光度法测定血浆微量亚甲蓝(MB)的方法。方法:利用丙酮沉淀血浆蛋白,硅胶G固相萃取纯化MB,分光光度法定量测定含量。结果:血浆中MB的回收率在20~100ng·mL^(-1)是96%~102%(RSD:2%~9%)。结论:本法可使血浆中MB得到很好的富集分离,萃取率较高且稳定。血浆试样的用量可以根据血药浓度的不同相应增减。方法简便,有较好的准确度和重现性,能够满足ng·mL^(-1)级血浆MB的分析需要。 Objective: A spectrophotometry method was developed for the determination of a trace level of methyl-ene blue (MB) in human plasma. Method: Plasma proteins were precipitated by acetone. MB was extracted and cleaned up on silica gel G solid - phase extraction cartridges. MB concentration was determined by spectrophotometry at 653 nm. Results: The average recovery of MB from human plasma at 20 - 100 ng·mL-1 were 96% -102 % ( RSD: 2% -9 % ). Conclusions: This method can separate and concentrate MB from human plasma efficiently, and the extraction rate was high and stable. The mass of plasma can be addition or reduction with the different concentration of MB in plasma. This method is simple, and has good accuracy and reproducibility. It is suitable for the determination of MB at ng · mL-1 level in human plasma.
