摘要
目的:寻找一种快速、有效的分离定量亚叶酸钙(LV)的方法。方法:采用高效液相色谱法,利用采用新方法合成的牛血清白蛋白手性固定相,以50 mmol·L^(-1)磷酸盐缓冲溶液(pH 6.5)为流动相对其进行拆分。结果:[6S]-、[6R]-LV分别在4.46 min和7.59 min时出峰,全过程可在10 min内实现基线分离,其分离度α=2.10,Rs=2.78。结论:流动相pH值、缓冲溶液浓度以及温度对[6S]-LV、[6R]-LV的容量因子都有较大的影响,并初步推测[6S]-、[6R]-LV与牛血清白蛋白之间是单个位点的作用。
Objective: A HPLC method was developed to completely separate [6S] -and [6R] -calcium folinate (LV) using BSA - CSP synthesized by a novel method. Method: To separate with 50 mmol · L-1 sodium phosphate buffers ( pH 6. 5) by a novel BSA chiral stationary phase. Result: Baseline separation was achieved in 10 min: 4. 46 min and 7. 59 min, a =2.10, Rs =2.78. Conclusion: [65] -LV, [6R] - LV's k' s were largely influenced by pH, concentration of mobile phase and column temperature, and it was presumed that LV possessed a single type of binding site on BSA.
