胰岛素样生长因子1对大鼠慢性压迫性脊髓损伤后神经功能恢复的影响 被引量:6

摘要 目的观察胰岛素样生长因子1(IGF-1)对慢性压迫性脊髓损伤后神经功能恢复的影响。方法50只Wistar雌性大鼠,随机分为正常组、慢性重度(60%)压迫组、减压组、生理盐水(NS)组及IGF-1组。正常组不做任何处理,其余各组置入平头塑料螺钉对大鼠脊髓进行后路渐进性压迫,于2个月后达到60%左右重度压迫程度,然后减压组、NS组和IGF-1组彻底减压,NS组和IGF-1组分别于减压后1、3、5、7、9d在蛛网膜下腔给予NS和IGF-1(25μl),并于减压后2、4、6、8、10d后肢运动功能评分和检测运动诱发电位(MEP)。结果压迫组大鼠后肢运动功能障碍,MEP波幅(P1~N1峰峰值)降低、潜伏期延长;IGF-1组大鼠后肢运动功能及MEP波幅及潜伏期有明显的恢复(P<0.05),且于4~6d后显著优于单纯减压组和NS组(P<0.05)。结论IGF-1可明显促进慢性压迫性脊髓损伤后大鼠后肢运动功能及MEP波形的恢复。 Objective To observe the effect of insulin-lik e growth factor-1o n motor function o f lower limbs and motor evoked potent ial(MEP)after chronic s pinal cord compression in rat.Methods 50Wistar rats were randomly divided int o normal group ,chronic compress ive group,decompressive group,NS grou p and IGF-1group.The animal model of chronic spinal cord compression was establishe d in all groups except for normal group.Compressiv e degree was achieved about 60%in two months.At the time ,decompression w as done in decompressive gro up,NS group and IGF-1group.NS or IGF-1were respectively supplied th rough subarachnoid cavity 1,3,5,7,9d after decompression in NS group and IGF-1group.Motor functio n of lower limbs and motor evoked pote ntial(MEP)were detected 2,4,6,8,10d after decompression.Results Motor function of l ower limbs and amp litude of motor evoked potential(MEP)reduced and delitescen ce of MEP prolonged in chronic compressive group.They obviously r ecovered in I GF-1group(P<0.05),and the recovery in IGF-1group was b etter than in decompressive group and NS group(P<0.05).Conclusions IGF-1can accele rate undee reversio n of MEP and promote motor function re covery of spinal cord after chronic compressive spin al cord injury.
