摘要
采用单细胞凝胶电泳技术研究了壬基酚(NP)对扇贝血淋巴细胞DNA的损伤作用。以0,0.025,0.10,0.25和1.00 mg/L的NP处理海湾扇贝24 h和48 h,检测其对血淋巴细胞DNA的损伤程度。结果表明,当NP在0.025 mg/L时就可以使扇贝血淋巴细胞产生轻微的DNA损伤,表现在DNA发生了拖尾,处理24 h后DNA拖尾率为10%,48 h后拖尾率为33%。随着NP浓度的增加和染毒时间的延长,血淋巴细胞的DNA拖尾率都相应增加,最高浓度的处理组(1.00 mg/L)中,DNA损伤最为明显,处理24 h后的拖尾率为80%,48 h后拖尾率为93%。该结果表明NP对扇贝血淋巴细胞具有遗传毒性损伤作用。
The effect of nonylphenol on DNA damage in scallop hemolymph cells was detected by single-cell gel electrophoresis assay. Scallops(Argopecten irradians) were exposed to various nonylphenol concentrations(0,0.025,0.10,0.25 and 1.00 mg/L) and DNA damage in hemolymph cells were detected after 24 and 48 hs.The results show that NP at 0.025 mg/L will cause slight damage to cellular DNA.The percentages of cellular damage are 10% and 33% after 24 and 48 hs,respectively.NP at 1.00 mg/L will cause serious damage to cellular DNA.The percentsages of cellular damage are 80% and 93% after 24 and 48 hs,respectively.NP,therefore,poses genetic toxicity to scallop hemolymph cells.
