钝顶螺旋藻多糖的分离纯化及psp2组分对HeLa细胞的影响 被引量:1

摘要 为研究钝顶螺旋藻多糖粗品中不同分子量多糖组分的抗肿瘤活性,应用Superdex-75凝胶纯化分离得到psp1,psp2,psp3和psp4 4个不同分子量多糖组分,分子量依次为65 400,16 900,14 400和11 700,其中psp2经Sephadex G-25凝胶层析柱脱盐后测得其糖含量为98.5%。psp2作用于体外培养的HeLa细胞,随其浓度增加,HeLa细胞存活率逐渐降低;流式细胞术证实psp2对肿瘤细胞存在周期特异性,使细胞发生G1期阻滞。 In order to investigate the anti-tumor activity of different molecular weight compositions from crude polysaccharides of Spirulina platensis(psp),Superdex-75 gel was employed to isolate and purify crude psp and four polysaccharide compositions with different molecular weights psp1,psp2,psp3 and psp4 were prepared,their molecular weights tested were 65 400,16 900,14 400 and 11 700,respectively,and the carbohydrate content of psp2 was 98.5% after salt removal by Sephadex G-25 gel filtration chromatography.In vitro experiment it showed that with the increase of psp2 concentration,the survival rate of Hela cells declined;Flow cytometric result proved that psp2 had a specific effect on the division cycle of Hela cells,increased the number of cells in phase G1.
