VEGFR-1,VEGFR-2 mRNA在局灶性脑缺血的表达及作用研究 被引量:3

摘要 目的 研究 VEGFR- 1 ,VEGFR- 2 m RNA在局灶性脑缺血的表达 ,探索二者的作用。方法 建立 SD大鼠永久大脑中动脉闭塞(MCAO)模型 ,应用原位杂交技术和免疫组化方法检测在局灶性脑缺血后不同时间点 VEGFR- 1 ,VEGFR- 2在 m RNA及蛋白质水平的表达。同时观察局灶性脑缺血后血管形成情况。结果 原位杂交显示 VEGFR- 1 ,2 m RNA在局灶性脑缺血后 3h即开始表达增强 ,于 48h达高峰 ,后 VEGFR- 1 ,2 m RNA逐渐下降 ,约在 3 w时达对照水平。免疫组化检测 VEGFR- 1 ,2在局灶性脑缺血后 6 h呈上升性表达 ,VEGFR- 1 ,2于 2 4 h达高峰 ,后在 2 w恢复到对照水平。二种方法均可见 VEGFR- 1 ,2主要在缺血周边血管内皮细胞表达 ,但也在神经元表达。在缺血后 48h半影区周边出现血管增生 ,并逐渐向中心区延伸 ,同时 VEGFR- 1 ,2的表达伴血管的增生 ,逐渐向缺血中心区移行。结论 在局灶性脑缺血早期 VEGFR- 1 ,2 m RNA在血管内皮细胞、神经细胞均有表达 ,对改善缺血半影区血供有重要作用。
