黄芪当归合剂对大鼠慢性肾硬化中骨桥蛋白表达的影响 被引量:31

摘要 目的 :探讨黄芪当归合剂对慢性肾硬化中骨桥蛋白表达的影响。方法 :采用大鼠慢性嘌呤霉素肾硬化模型 ,随机分为模型组 ,黄芪当归合剂治疗组 (简称中药治疗组 ) ,血管紧张素受体拮抗剂 (Irbesar tan)治疗组 (简称AT1RA组 )。实验全程 12周 ,血、尿标本进行生化检测、肾组织病理染色和免疫组化检测骨桥蛋白 ,单核巨噬细胞及细胞外基质层粘连蛋白 (laminin)、核心蛋白聚糖 (decorin)的表达。结果 :黄芪当归合剂具有与Irbesartan相似的减少尿蛋白 ,改善肾功能的作用 ;同时肾组织病理损伤减轻 ,尤其是小管间质损伤指数明显降低 ;间质细胞外基质 (decorin和laminin)聚集明显减少 ;小管骨桥蛋白的表达和间质单核巨噬细胞浸润均明显减少 ,且两者呈正相关性 (r =0 885 ,P <0 0 1)。结论 :黄芪当归合剂具有与Irbesartan相似的肾脏保护作用 ,这一作用可能涉及骨桥蛋白表达上调的抑制。 Objective: To observe the effect of Astragalus Angelica Mixture (AAM) on osteopontin (OPN) expression in rats with chronic nephrosclerosis. Methods: Chronic nephrosclerosis model rats induced by repeated intraperitoneal injection of puromycin were randomly divided into the model group, AAM group and Irbesartan (an antagonist of angiotensin) group. The experimental course lasted 12 weeks. Blood and urine samples were examined by biochemical method. Kidney tissue was taken for pathological stain and immunohistochemical method and was applied to examine OPN expression, mononuclear macrophage, laminin in extracellular matrix and decorin expressions. Results: AAM showed the effects of decreasing urinary protein and improving renal function similar to that of Irbesartan. It also could alleviate the pathological damage of kidney tissue, especially in decreasing renal tubular mesenchymal damage index. The accumulation of decorin and laminin in the mesenchymal extracellular matrix significantly decreased. Renal tubular OPN expression and mesenchymal infiltration of mononuclear macrophage decreased significantly and in a positive correlated manner (r=0.885, P<0 01). Conclusion: AAM has similar renal protective action to that of Irbesartan, this action may be related to the inhibition of up regulated OPN expression.
