He-Ne激光宽束照射对亚甲兰敏化HeLa癌细胞的生物学效应

摘要 本文报道了浓度为5μg/ml亚甲兰对HeLa癌细胞的光动力学杀伤作用。结果表明,经亚甲兰敏化的HeLa细胞,在4.42J/cm^2或5.7 J/cm^2两种剂量的He-Ne激光宽光束照射下,其存活细胞数随着激光照射剂量的增加而减少;其显微结构及超微结构也发生明显变化:细胞变圆,膜表面微绒毛减少,胞质内产生大量空泡,线粒体也受到一定程度损伤。此外,用组织化学方法也观察到其ATP酶的活性受到明显抑制。 The experimental results about the photodynamic effect of He-Ne laser irradation on HeLa cell sensitized by methylene blue (MB) with the concentration of 5μg/ml are obtained. It shows that the number of survival cells sensitized by MB is decreased with the increase of irradiation dose, and the growth rate of cell also decreases obviously when the energy density of He-Ne laser is 4.42J/cm^2 or 5.73 J/cm^2. In comparsion with the control group, the changes in the microstr- ucture and ultrastructure of the cell sensitized by MB and irradiated with He-Ne laser of 5.73 J/cm^2 have taken place, which include the decrease of microvilli on cellular surface, the increase of vacuoles in cytopalsm and swelling of mitochondria. Furthermore, the activity of ATPase is inhibited significantly.
