大鼠原位性肾小球肾炎模型的建立 被引量:13

A NEW MODEL OF EXPERIMENTAL GLOMER- ULONEPHRITIS IN RATS BY ADMINISTRATION OF CATIONIC BOVINE SERUM ALBUMIN

摘要 应川阳离子化牛血清白蛋白(C-BSA)诱导Wistar大鼠产生膜性肾炎,结果表明:经静脉注射C-BSA2.5mg第2周始出现严重蛋白尿,第3周进一步加重,第4用肾脏病理检查可见GBM增厚,钉突形成,上皮下免疫沉积物及足突损伤。该模型发病率为100%,重复性好,具有人类膜性肾炎的病理炎型特点和肾病综合征的临床表现,而无肾功能损害,是肾炎的发病机理和药物防治研究的理想动物模型。 The study developed a new model of experimental glomerulonephritis in rats by administration of cationic bovine serum albumin(C-BSA) as antigen. Twenty-four male wistar rats, weight 100~150g, were at random divided into two groups. One group(n=11) was as control; preimmunized by native bovine serum albumin(N-BSA) 1 mg and Freund's incomplete adjuvant(FIA) subcutaneously 7 days before starting injection. And another group (n- 13) was as experimental preimmunized by C-BSA 1 mg and FIA. Then the rats or both groups were respectively given intravenous injection of N-BSA or C-BSA 2.5mg thrice weekly for 3 weeks. At the end of 4th week, all rats were killed. All rats given C-BSA had been induced heavy proteinuria and nephrotic syndrome since 2nd week, but the rats given N-BSA showed slighter proteinuria only. Renal lesions produced by C-BSA were markedly different from that found in rats given N-BSA, and showed subepithelial deposits, extensive fusion of foot process, thickness and prominent spikes of GBM and mild mesangial proliferation, whereas rats receiving N-BSA seemed to be normal. This model showed several advantages, such as being reproducible, consistent, convenient and inexpensive, especially having heavy proteinuria and nephrotic syndrome without impairing renal function. These fat features make the model eminently suitable for further study.
