摘要
通过对烤烟 K32 6不同外植体、不同培养基及影响烟草体细胞胚发生因素的研究。结果表明 :烤烟 K32 6产生愈伤组织的百分率因不同外植体而异 ,在含 2 ,4 - D 1.0 mg/L的 MS固体培养基上产生愈伤组织的百分率分别为 5 2 %、2 6 %、4 % ,而在含 NAA 0 .1mg/L和 KT 2 .0 mg/L的 MS去分化培养基上产生愈伤组织的百分率分别为 6 3%、4 4 %、5 % ;体细胞胚发育是不同步的 ;一定浓度的 2 ,4 - D对烤烟体胚发生是必须的 ,但高浓度 2 ,4 - D将抑制体胚的形成 ;2 %蔗糖比 3%蔗糖利于烤烟 K32 6体胚发生。
Study on the influence of media, factors and explant on the percentage of callus formation for flue cured tobacco K326. It showed that the percentage of callus formation for flue cured tobacco K326 was different, in variant explants. The percentage of callus formation was respective 52%,26% and 4% in MS media containing, 2,4 D 1.0mg/L, it was respective 63%,44% and 5% in MS polarication media containing NAA 0.1mg/L and KT 2.0mg/L and KT 2.0mg/L. The development of somatic embryos was in different stages; The suitable degree 2,4 D was essential to developing flue cured tobacco of somatic embryos, while high degree 2,4 D would restrain flue cured tobacco of somatic embryos.2% saccharose was much better than 3% on development flue cured tobacco K326 of somatic embryos.
