摘要
对琼脂单、双扩散法在牛初乳IgG测定中的应用进行分析 ,建立的单扩散标准曲线为y =17.193x +8.0 0 5 ,γ =0 .9913,板间变异系数 2 .8%~ 5 .5 % ,板内变异系数 2 .6 3%~ 5 .33% ,回收率 87.0 %~ 12 7.0 %。双扩散中标准牛IgG标记为效价 1∶2 2 2 2 .2 ,批内变异系数 0 %~ 2 2 .2 % ,批间变异系数 18.8% ,回收率 87.5 %。统计分析表明两种方法测定值无显著差异 (α =0 .0 5 )。相比而言 ,单扩散法精密度好 ,双扩散方法更简便 ,抗体试剂用量为单扩散法的 2 %~ 5 % ,检测成本低。
The applications of agar single/double immunodiffusion method in determination of immunoglobulin in bovine colostrum were analysed,the established curve of agar single immunodiffusion method was y=17.193x+8.005 r=0.9913,the coefficient of variation among plates was 2.8%-5.5%,whereas coefficient of variation in plates was 2.63%-5.33%,the recovery rate was 87.0%-127.0%.The potency of IgG standard in agar double immunodiffusion was 1:2222.2,the coefficient of variation in group was 0-22.2%,whereas coefficient of variation among group was 18.8%,the recovery rate was 87.5%.The statistical analysis showed that there were no significant difference between results obtained from the two methods (a=0.05).The agar single immunodiffusion was more precise as compared with the agar double immunodiffusion,however the latter was simple and low-cost with the reagent consumption being 2%-3% that of the former one.Enterprises could make choices based upon their own demands.
