Quantitative analysis of HCV-RNA of different genotypes

摘要 目的 根据定量 PCR检测结果 ,了解血清不同基因型 HCV- RNA的含量 .方法 采用荧光定量 PCR和逆转录巢式定性 PCR同时检测 16 6例丙肝患者的血清 HCV- RNA,再进行酶切分型 .计算各型 HCV- RNA平均含量 .结果 定量 PCR阳性血清 (131例 ) HCV- RNA含量在 1× 10 4 .0 4~ 1×10 8.1 1拷贝· m L- 1 .HCV- 型 (10 6例 )、HCV- 型 (18例 )和 / 混合型 (3例 ) RNA平均滴度分别为 1× 10 6 .88± 2 .0 1拷贝· m L- 1 ,1× 10 5.79± 1 .82 拷贝· m L- 1 和 10 5.50± 1 .6 2 拷贝· m L- 1 .结论 定性和定量检测结果有很好的一致性 .HCV- 型病毒含量显著高于 HCV- 型 . AIM To investigate the quantity of HCV RNA of various genotypes in sera through the results of quantitative PCR. METHODS HCV RNA was detected in 166 hepatitis C patients using quantitative PCR and reverse transcription nested polymerase chain reaction (RT nested PCR). HCV RNA positive sera were genotyped by the restriction fragment length polymorphism (RFLP) analysis of PCR productsfrom 5NC region. The mean amount of HCV RNA was counted in various genotypes. RESULTS The amount of HCV RNA ranged from 1×10 4.04 copies·mL -1 to 1×10 8.11 copies·mL -1 in quantitative PCR positive sera (131 cases). The mean titer of HCV RNA in HCV Ⅱtype (106 cases), Ⅲ type (18 cases) and Ⅱ/Ⅲ mix type (3 cases) were 1×10 6.88±2.01 copies·mL -1 , 1×10 5.79±1.82 copies·mL -1 and 10 5.50±1.62 copies·mL -1 , respectively. CONCLUSION The results of qualitative PCR and quantitative PCR detection were similar. The serum HCV RNA concentration in patients with HCVⅡ infection was significantly higher than that in patients with HCV Ⅲ infection.
