摘要
目的 硝普钠 (SNP)对高血压病 (EH)患者动脉血管平滑肌细胞 (VSMC)的一氧化氮合酶 (NOS)活性和一氧化氮(NO)含量、细胞周期、细胞数量、细胞增殖指数的影响 .方法 对血压正常 (NT)者和 EH患者的肠系膜动脉进行分离培养 ,对比观察在基础状态和 SNP作用下 ,EH组和 NT组动脉VSMC的 NOS活性、NO含量、细胞周期、细胞数量、细胞增殖指数的变化 ,从而研究 SNP对 EH患者的动脉 VSMC的NOS- NO系统及细胞增殖的影响 .结果 1在基础状态下 ,EH组 VSMC的 NO含量、NOS活性低于 NT组 ,而且两组间有显著差异 (P<0 .0 5 ) ;2在 SNP作用下 ,EH组 VSMC的NO含量、NOS活性较 NT组减少 ,但两组间无明显差异 (P>0 .0 5 ) ;3在基础状态下 ,EH组动脉 VSMC的 S期的百分率、细胞增殖指数 (PI)、细胞数量均明显高于 NT组 ,且两组间有明显差别 (P<0 .0 5 ) ;4在 SNP作用下 ,EH组动脉 VSMC的S期的百分率、细胞增殖指数 (PI)、细胞数量均高于 NT组 ,且两组间无明显差别 (P>0 .0 5 ) .结论 NO的供体药物 SNP可以通过分解产生大量的外源性 NO,纠正 EH患者 VSMC的 NO含量不足的缺陷 ,抑制 EH患者动脉 VSMC的增殖 ,从而抑制 EH患者动脉血管的重构 ,阻止 EH的发生、发展 .
AIM To investigate the change in NOS activity, NO content, cell number, cell proliferation in essential hypertention (EH) and normotension (NT) under nitriprusside impact and basical condition. METHODS Mesenteric artory VSMC of the EH and NT groups were cultured and the changes in NOS activity, NO content, cell number, cell proliferation index were observed in order to explore the varity of NO content, cell number, cell prolilferation index (PI) of EH and NT group in SNP and basical condition. RESULTS ① Under badsical condition, the NO content and NOS activity of EH group is lower than that of NT group and the difference between them is obvious ( P <0.05). ② Under SNP influence , the NO content and NOS activity of EH group is lower than that of NT group, difference between them is not obvious ( P >0.05). ③ Under basical condition, the cell number and cell proliferation index of EH group is higher than that of NT group and the difference between them is obvious ( P <0.05). ④ Under SNP impact, the cell number and cell proliferation index of EH group are higher than that of NT group and the difference between them is not obvious ( P >0.01). CONCLUSION SNP can improve NO content of EH group and inhibit cell proliferation of EH. Therefore in the early stage of EH, we can use SNP to stop the development of EH.
