摘要
目的 探讨抑癌基因 p2 7与原发性肾细胞癌 (prima-ry renal carcinoma,RCC)发生及恶性程度的关系 .方法 应用免疫组织化学方法检测了 86例 RCC及癌旁组织中 p2 7蛋白的表达 .结果 p2 7蛋白免疫组化阳性信号为棕黄色颗粒 ,主要分布在胞质 ,少数细胞核内也可见阳性显色 ,p2 7蛋白在癌组织及癌旁组织中的阳性率分别为 33.7% (2 9/ 86 )和 86 .4 % (74 / 86 ) ,两者比较有显著性差异 (P<0 .0 1) . 86例 RCC标本中 , 级、 级和 级中 p2 7蛋白的检出率分别为 5 0 .0 %(2 2 / 4 4) ,2 6 .3% (5 / 19)和 8.0 % (2 / 2 3) ,p2 7蛋白在高分化RCC中的表达率明显高于低分化 RCC组 (P<0 .0 5 ) .结论 抑癌基因 p2 7的表达和分布与 RCC的发生、发展及恶性程度密切相关 ;p2 7蛋白的表达低 ,肿瘤分级高 。
AIM To study the expression of p27kip1 in renal carcinoma and its relationship with tumor grade. METHODSThe expression of p27kip1 was detected by immunohistochemical method in 86 cases of renal carcinoma and 86 cases of paracarcinoma. RESULTS The brown color was detected by immunohistochemistry in positive cells. Within tumor cells, the p27kip1 protein staining was predominantly cytoplasmic and less intense nucler staining was also observed. The positive expression rate of p27kip1 protein was 33.7% (29/86) in tumor and 86.4% (74/86) in paracarcinoma. There was a significant relationship betwent them ( P < 0.01 ). In all the cases, 44 cases were gradeⅠ, 19 cases gradeⅡ and 23 cases gradeⅢ. The positive expression rate of p27kip1 protein was respectively 50.0%, 26.3% and 8.0%. p27kip1 expression was strongly associated with tumor grade in renal carcinoma ( P <0.01). CONCLUSION The results suggest that the expression of p27kip1 protein in renal carcinoma showed a siginficant relationship with tumor grade. p27kip1 protein is frequently expressed at low level in poorly differentiated tumors. Decreased p27kip1 protein staining correlated with higher tumor grade.
