摘要 目的 用表达端粒酶反义 RNA的方法研究端粒酶在喉癌细胞生长中的作用 .方法 将端粒酶反义 RNA真核表达载体 p BBS2 12 - h TR转染入人喉癌 Hep- 2细胞系中 ,采用端粒重复扩增法测定端粒酶活性 ,用 MTT法及流式细胞仪检测细胞周期 ,并用电镜观察细胞生长状况 .结果 端粒酶反义 RNA表达后能抑制 Hep- 2细胞的端粒酶活性 ,抑制细胞增殖并促进其凋亡 .结论 以端粒酶反义 RNA降低端粒酶表达的方法对人喉癌细胞系 Hep- 2的生长有明显抑制作用 . AIM To study the effect of telomerase on Hep 2 cells by telomerase antisense RNA expression. METHODS Using lipofectin mediated DNA transfection, telomerase antisense RNA was successfully transfected into Hep 2 cells. By means of TRAP, MTT, FCM and electromicroscope, the changes in Hep 2 cells in telomerase activity, cell cycle and dynamics of cell growth were observed. RESULTS Telomerase antisense RNA significantly inhibited telomerase activity and the proliferation of Hep 2 cells, and accelerated its apoptosis. CONCLUSION Telomerase antisense RNA repressed expression can inhibit laryngeal carcinoma cells of Hep 2 growth significantly.
