摘要
目的 ,为pSVPoMcat微基因修饰雪旺氏细胞 (SC)移植对实验性脊髓损伤 (SCI)的治疗作用提供可靠的形态学依据。方法 :采用切割法制备脊髓半横断损伤模型 (T8平面 ) ,实验动物随机植入pSVPoMcat微基因修饰的SC组 (A组 )、SC组 (B组 )和明胶海绵组 (C组 ) ;每组 10只 ,采用定量酶组织化学方法比较A、B、C三组脊髓前角细胞内乙酰胆碱酶 (AchE) ,酸性磷酸酶 (ACP)活性 ;应用定量免疫组织化学方法比较脊髓白质内轴突数量。结果 :(1)损伤后不同时间A、B、C组前角细胞AchE活性为A <B <C ,而ACP活性为A >B >C ;(2 )损伤后脊髓白质一定面积内轴突数量为A >B >C。结论 :pSVPoMcat微基因修饰雪旺氏细胞移植对实验性脊髓损伤 (SCI)
Objective:To provide reliable morphological evidence of the therapeutic effects of microgene pSVAoMcat implanted to modify the schwann cells(SC)on the experimental spinal cord injury(SCT).Methods:Experimental animals were divided into three groups∶the group of microgene pSVPoMcat implanted to genetically SC(group A),SC implanted group(group B)and the control group(group C),each group having 10 animals.Acetylcholinesterase(AchE)activity of anterior horn cells in the A,B,C groups were measured respectively with the quantitative enzyme histochemistry,The number of axon in the spinal the white matter was surveryed in the three groups by means of the quentitative immunohistochemistry.Results∶1.AchE activity of anterior horn cells in the A,B,C groups were A<B<C at varivous times,and ACP activity were A>B>C;2.The number of axon in a certain area of spinal the white matter in the A,B,C groups were A>B>C,Conclusion:The therapeutic effects of microgene pSVPoMcat implanted to modify SC on the experimental SCI have the reliable morphological evidence.
