黄芪和药饼灸对5-Fu治疗中所致荷结肠癌小鼠免疫功能低下的正向调节作用 被引量:2

摘要 目的 研究黄芪、药饼灸对经 5 FU抗瘤治疗引起免疫功能低下小鼠的正向调节作用。方法C2 6小鼠结肠癌细胞株 5× 10 5于BALB c小鼠皮下注射建立小鼠结肠癌实验模型。接种肿瘤细胞后 2 4h开始治疗实验。实验共分 5组 ,分别为生理盐水组 (A组 ) :即穴位 (足三里 )注射生理盐水 0 .1mL ,隔d 1次 ,共 2wk ;5 FU治疗组 (B组 ) :腹腔注射 5 FU 0 .2 5mg 0 .1mL(12 .5mg kg) ,每隔 2d 1次 ,共2wk ;5 FU联合黄芪组 (C组 ) :腹腔注射 5 FU 0 .2 5mg 0 .1mL(12 .5mg kg) ,每隔 2d 1次 ,同时隔d穴位 (足三里 )注射黄芪注射液 0 .1mL(2 g mL) ,共 2wk ;5 FU联合药饼灸组(D组 ) :腹腔注射 5 FU 0 .2 5mg 0 .1mL(12 .5mg kg) ,每隔 2d 1次 ,同时中腕、神阙、关云穴位用药饼灸 ,每穴灸 3壮 ,隔d 1次 ,共 2wk ;5 FU联合黄芪加药饼灸组 (E组 ) ;治疗方法同上。治疗结束后 ,获取小鼠脾脏单个细胞 ,分别采用流式细胞术 (FACS)测定脾淋巴细胞中T细胞亚群及NK细胞表面标志。采用 4hLDH释放法 ,检测小鼠淋巴细胞 (NK、CTL)的杀伤功能。结果本实验结果表明 ,药饼灸加黄芪联合 5 FU组的上述免疫指标 (包括CTL、NK细胞的杀伤活性 )与 5 FU治疗组相比较具有统计学意义。结论黄芪注射液加药饼灸联合 5 FU ,对单经 5 FU? Aim To study on positive immuno-regulation of Huangqi and Yao Bingjiu on the mice bearing inoculated colon cancer cells treated with 5-Fu. Methods An experimental model was established by hypodermic inoculating the mouse colon cancer cell lines C26 of 5×10 5 into the Balb/c mice. The experimental treatment was given to the injected mice 24 hrs after inoculation. 5 groups were set up in the study. Group A: NaCl solution was injected to the acupoint Tsusanli, every other day for 2 weeks. Group B: 12.5 mg/kg of 5-Fu was injected to the peritoneal cavity on the mice, every two days for 2 weeks. Group C: 0.1mL of Huangqi solution was injected to the acupoint of Zusanli every other day for 2 weeks, jointly with the same 12.5 mg/kg of 5-Fu was injected to the peritoneal cavity every two days for 2 weeks. Group D: 12.5 mg/kg of 5-Fu was injected to the peritoneal cavity, meanwhile the acupoint of Zhongwan, Shenque and Guanyun were treated with Yao Bingjiu. 3 moxa-cones were used to every acupoints, every other day for 2 weeks. Group E: The mice were treated with 5-Fu as Group B, combined with treatment of Huangqi injection and Yao Bingjiu as Group C and Group D respectively. The mouse spleens were taken out and the monocytes were isolated. The cells were labeled by direct immuno fluorescence. The T cell subsets and CD antigens expressed on NK cells were analyzed by FACS. The cytotoxicities of NK cells and CTLs were detected by LDH release assay after coincubated with target cells. Results The cytotoxicities of NK cells and CTLs from groups C, D and E were higher than those of mice only treated with 5-Fu. Conclusion Huangqi and Yao Bingjiu can upregulate the immune function of mice bearing colon cancer.
