摘要 目的 探讨KAI1基因在胰腺癌中的突变情况。方法 对17例伴有淋巴结转移的原发性胰腺癌(Ⅲ期),7例无淋巴结转移的原发性胰腺癌(2例Ⅰ期,5例Ⅱ期)和9例正常胰腺组织标本进行KAI1基因突变分析。结果 24例胰腺癌标本中,7例证实有KAI1点突变。这种突变出现在886位核苷酸上,导致从A到G的转变,缬氨酸置换异亮氨酸,且有突变的癌标本均属于晚期有转移的胰腺癌(Ⅲ期)。结论 上述结果提示KAI1基因突变使KAI1 mRNA水平降低可能是胰腺癌转移的主要因素之一。 Objective To detect mutations of the KAI1 gene in pancreatic cancer. Methods 17 primary pancreatic cancer samples with lymph node metastases( stage II), 7 primary pancreatic cancer samples without metastases (2 in stage I ,5 in stage II ) and 9 normal pancreatic tissue samples were analyzed for KAI1 mutations. Results Point mutation was found in 7/24 pancreatic cancer samples (29%), all mutations located at nucleotide 886 which resulted in an A to G transition and an amino acid switch from isoleotide to valine. All cancer samples with mutations belong to metastatic tumors ( stage III). Conclusions KAI1 gene mutations occur frequently in advanced pancreatic cancers which might contribute to the decrease of this gene.
