外源一氧化氮对大鼠胰岛细胞损伤机制研究 被引量:14

摘要 目的:研究外源NO对胰岛细胞的损伤机制和功能的影响。方法:采用DNA电泳技术以及单细胞电泳检测NO对胰岛细胞的凋亡和DNA的损伤作用;通过测定胰岛素含量检测胰岛细胞在NO作用下的合成和分泌功能。结果:外源NO能导致胰岛细胞DNA的断裂形成彗星细胞,DNA电泳显示典型梯状条带,大剂量NO形成死亡条带;同时NO能抑制胰岛细胞合成和分泌胰岛素。结论:外源NO可导致胰岛细胞凋亡、DNA断裂损伤并抑制其合成分泌胰岛素的功能。 <Abstrcat>Objective:To study the mechanism of the destruction and changes of function of islet cells caused by exogenous nitric oxide(NO).Methods:The damage of DNA and apoptosis in islet cells were assessed in vitro with single cell gel electrophoresis and gel electrophoresis of DNA. Insulin synthesis and secretion of normal rats pancreas under high level ofglucose caused by NO were detected.Results:The exogenous NO caused breakage of DNA strand .The islet cells showed remarkable apoptotic ladder with low dose of NO and necrosis ladder with high dose.The insulin synthesis and secretion of islet cells under high level of glucose were inhibited by NO in vitro.Conclusion:The exogenous NO can directly induce islet cells apoptosis,DNA breaking and inhibit their function.
