米诺环素对2型糖尿病大鼠海马中PAI-1表达的影响

摘要 目的:探讨纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor type-1 PAI-1)在2型糖尿病大鼠海马中的表达情况以及米诺环素对PAI-1表达的影响。方法:高糖高脂饮食加小剂量链脲佐菌素造2型糖尿病模型,将SD大鼠随机分为正常组、糖尿病组、米诺环素组,给药两周后测各组大鼠海马中PAI-1表达情况。结果:PAI-1在正常组大鼠海马C3区有很弱表达,在糖尿病组表达显著增强(P【0.01),在米诺环素组表达比糖尿病组显著减弱(P【0.01)。结论:2型糖尿病大鼠海马神经元内PAI-1的表达异常增高,米诺环素可减少PAI-1的表达,这可能是米诺环素保护糖尿病认知功能障碍的机制之一。 Objective:To explore the expression of plasminogne activator inhibitor type – 1(PAI-1)in the hippocampus in a type 2 diabetes model of rats and the change after the treatment with minocycline.Methods:A model of type 2,diabetes mellitus was induced by a high-glucose and high-fat diet and low doses of STZ.Rats were then randomly divided into 3 groups:control group,DM group and minocycline group.After twe-week's treatment with minocycline,examine the expression of PAI-1 in the rats' hippocampus of every group.Results:PAI-1 was weakly expressed in the control group.The expression was significantly increased in the DM group(P<0.01).While the expression was noticeably decreased in the minocycline group in comparation with the DM group.Conclusion:the expression of PAI-1 was significantly increased in the hippocampal neuron of a type 2 diabetes model of rats.Minocycline could reduce the expression of PAI-1,which could be one of the mechanisms of minocycline to protect the impaired cognition caused by diabetes.
