转化生长因子β1及其受体在肝胆管结石并发肝胆管癌中表达的意义

Study the relationship between the expression of TGF-β1、TGF-βR-II and carcinogenesis of cholangiocarcinoma associated with hepatolithiasis

目的:探讨TGFβ1及其受体TGFβRII在肝胆管结石并发肝胆管癌过程中所起作用。方法:应用免疫组化S-P法检测TGFβ1及其RII在肝胆管结石症及结石合并肝胆管癌组织中表达。结果:在肝胆管结石胆管上皮及腺体组织不同类型病变各组中,TGFβ1阳性分级表达呈:正常<单纯增生<不典型增生<癌,P<0.01;TGFβRII阳性分级表达呈:正常>单纯增生>不典型增生>癌,P<0.01。肝胆管癌组织中TGFβ1、TGFβRII表达与癌分化程度无明显相关,P>0.05。结石合并肝胆管癌组织中TGFβ1与TGFβRII表达呈负相关,P<0.01。肝胆管癌组织中TGFβ1与Ki67表达呈正相关,TGFβRII与Ki67表达呈负相关,P<0.05。结论:在长期结石及炎症的刺激下胆管上皮细胞低表达TGFβRII导致TGFβ1无法发挥其生长抑制作用在结石致癌过程中有重要意义。

Objective:Investigate the relationship between the expression of and TGF-β1、TGF-βRII carcinogenesis of cholangiocarcinoma associated with hepatolithiasis.Methods:Immunohistochemistry(S-P) was used to detected TGF-β1、 TGF-βRII and Ki67 antigen.Results:The expression level of TGF-β1 in normal and abnormal biliary epithelium and grand of hepatolithiasis showed a step-wise increase in following order:normal<hyperplasia<atypical hyperplasia<carcinoma,P<0.01.The expression level of showed a step-wise decrease in following order:normal>hyperplasia>atypical hyperplasia>carcinoma,P<0.01.The expression of bFGF、FGFR-I was not significantly related to histological differentation of cholangiocarcinoma(P >0.05).The expression TGF-β1 of was in negative correlation with TGF-βRII in cancerous tissues(P<0.01).The expression of TGF-β1 was in positive correlation with Ki67 antigen in cancerous tissues;The expression of TGF-βRII was in negaitive correlation with Ki67 antigen in cancerous tissues;(P<0.01).Conclusion:TGFβ1 can`t act as its function of growth inhibition caused by low expression of TGFβRII in biliary epithelium cell is an important step in carcinogenesis of cholangiocarcinoma associated with hepatolithiasis.
