摘要
研究了非水介质毛细管电泳 -安培检测 (NACE -AD)罗红霉素的方法,着重探讨了非水介质、支持电解质、酸碱度和电极电位对分离测定的影响;以铂丝为检测电极 ,电极电位为0.93V ,在以甲酰胺 -甲醇(体积比1∶1)为有机溶剂 ,10mmol/L乙酸铵 -2mmol/L乙酸为缓冲液的非水体系中 ,并用样条小波最小二乘法进行数据处理 ,罗红霉素的检出限为0.1mg/L,在5~100mg/L范围内的线性回归方程为 :S=80.1+73.1ρ,r=0.9994;用该法检测罗红霉素制剂 ,回收率为94%~96% 。
A method for the determination of roxithromycin by non-aqueous capillary electrophoresis(NACE) with amperometric detection(AD) is presented. Several parameters affecting the NACE-AD determination were studied, including composition of non-aqueous media, acidity and concentration of running buffer, and potential of working electrode. The obtained data were modified by spline wavelet least square. Roxithromycin was well separated with other ingredients of roxithromycin tablets and roxithromycin dispersible tablets in a non-aqueous system of 10 mmol/L NH4Ac-2 mmol/L HAc buffer in formamide-methanol(1∶1 by volume) mixed solvents at platinum working electrode potential of 0.93 V. A linearity exists between peak area(S) and mass concentration of roxithromycin(ρ) in the range of 5~100 mg/L with regression equation S=80.1+73.1 ρ(r=0.999 4). Detection limit of 0.1 mg/L was obtained. The method was satisfactorily used for the analysis of roxithromycin preparations with recoveries of 94%~96%.
