摘要 目的采用骨髓间质干细胞体外转化为心肌细胞 ,为心衰的干细胞移植治疗提供新的细胞材料。 方法抽取贵州香猪骨髓液 3ml,按照Wakitani的方法培养出骨髓间质干细胞 ,经 5 -氮胞苷刺激 2 4h后 ,进行结蛋白、肌球蛋白重链、心肌特异性肌钙蛋白Ⅰ、连接蛋白 - 4 3免疫组化 ,电镜鉴定。 结果骨髓间质干细胞经 5 -氮胞苷刺激后 ,部分细胞呈梭形 ,结蛋白、肌球蛋白重链、心肌特异性肌钙蛋白Ⅰ、连接蛋白 - 4 3免疫组化阳性 ,电镜示梭形细胞有明显的肌丝 ,细胞核呈单椭圆形 ,位于细胞中央。 结论骨髓间质干细胞可在体外的环境下向心肌细胞转化 。 Objective To find out in a trial the potential capability of stem cells transplantation for heart failure. Methods Three milliliters of bone marrow were extracted from the posterior superior iliac spine of the Gui-zhou Xiang pork. Mesenchymal stem cells were cultured according to the Wakitanis method. After being co-cultured with 5-azacytidine for 24h, the cultured cells were evaluated by transmission electron microscopy and immunohistochemical staining for myosin heavy chain(MHC), desmin, cardiac specific troponin I(c TnT I) and connexin-43(Cx-43). Results After being co-cultured with 5-azacytidine, some MSCs became spindle-like. These cells were found positively stained with MHC, desmin, c TnT I and Cx-43. Transmission electron microscopy showed that myofilaments were formed with oval nuclei lying in the center of cells. Conclusion Cardiomyocytes can be generated from MSCs in vitro. This suggests that MSCs might be a new cell source for stem cells transplantation in heart failure therapy.
